Fluorescent Microscopy of Uterine Tissue
The objectives of this work are to describe, through the use of fluorescent microscopy analysis, a specific type of uterine tissue response to external stimuli. The main purpose of this experiment was targeting certain types of cells within the structural components of the uterus through immunofluorescence approaches. Fluorescence microscopy involves applying fluorescent staining to various cellular substructures and screening them.
Among the reproductive organs, the uterus contains distinctive layers assigned with specific functions, such as the endometrium and myometrium. Our focus was to label the epithelial cells that line the endometrium and the smooth muscle cells present in the myometrium with fluorescent tags to show where these are distributed. We were able to see cellular detail that provided a fuller view of how these layers work together to affect the functioning of the uterus.
My hypotheses were as follows:
Hypothesis 1: A fluorescent marker targeting the epithelial cells will bind to the endometrial layer, showing strong fluorescence.
Prediction: This would localize in the epithelial cells, and the marker would be giving a strong signal of fluomorphism in the endoplasmic membrane layer.
Hypothesis 2: One more of the experiment markers that get to smooth muscle and, consequently, bonds to the myometrial layer where it was used.
Prediction: The fluorescent signal will be created within the smooth muscle cells to be the cause of the development of the entire structure in the myometrium.
These cell types have been localized within the uterine tissue by use of fluorescent microscopy. Through the use of immunofluorescent markers, the epithelial cells of the endometrium and the smooth muscle cells in the myometrium are seen in great detail regarding their distribution and structure.
To ensure that the binding of markers would occur specifically, a parallel control was prepared by not adding the primary antibody. The control indeed expressed minimal fluorescence, while the experimental sample from direct fluorescence with antibodies specific for cytokeratin and smooth muscle myosin heavy chain did, in fact, result in fluorescence in targeted regions: epithelial and smooth muscle cells in their respective layers.
