Propose two different models that explain how the auroraB kinase is turned off when
amphitellic microtubule attachment is achieved. By “turned off” I mean that it stops
phosphorylating NDC80 and other kinetochore microtubule binding proteins.
Unphosphorylated NDC80 has a slower off rate from microtubules than phosphorylated
NDC80 so turning off auroraB stabilizes correct microtubule attachments. Your models
do not have to be original. You can use models shown in Marsoner et al. but you may
use ideas from other papers or come up with your own original idea. Original cartoon
drawings are encouraged. Please do not screen shot Fig 5 into your paper. Carefully
explain how your two models differ from each other and explain clearly whether they are
mutually exclusive or not. Citations for statements of fact in section 1 can be review
articles or primary research papers but cannot by websites or class lectures. 20 pts. 10 pts
for each model. If a model is incomplete, eg. CPC translocates from the outer
kinetochore to the inner centromere without suggesting how this only happens under
tension, 6/10 for that model.
2- Review the published literature that supports each model. In this section, clearly
describe each experiment and result and how the result supports one model. For each
result, clearly explain whether the result supports one model and excludes the other
model or if it only addresses one model. For section 2, cite only original research
articles, not review articles. You may cite experiments from Marsoner et al. as well as
any other paper you find. 20 pts. 10 pts/model
3- Propose one new experiment that would test model 1 and one new experiment that
would test model 2. Marsoner et al. make it very clear that your models can likely only
be tested in very specific genetic backgrounds. Clearly explain the genetic background
for each experiment.
20 points. 10 pts for experiment testing each model.
Your paper will be 3 to 5 pages of 12 pt font, single spaced, not including references or
cartoon drawings.
Spelling and grammar are expected to be perfect. All statements of fact in section
2 should be referenced to a primary research paper (not to a review article). Please
reference citations the way they are cited in your reading assignments.
Use the past tense to describe the procedures and results in published papers because
these experiments occurred in the past. Use the present tense only to describe
conclusions that are accepted facts today. Proposed experiments should be described in
the future tense or future perfect tense
